A UV-Vis spectrophotometer is a very common tool in a chemist's toolbox. This versatile measurement technique has a secure place in the chemistry laboratory.
A spectrophotometer, also known as a UV-Vis spectrometer, is a device for measuring the amount of a substance based on its electromagnetic absorption. Most of the substances analyzed in the spectrometer are in liquid form. Light is emitted in a visible spectrophotometer using a tungsten lamp, and in a UV spectrometer using a deuterium lamp. A spectrophotometer is used for the qualitative and quantitative measurement of a wide range of substances in various scientific and research fields such as chemistry, biochemistry, pharmacy, materials and the environment.
How a UV-Vis Spectrophotometer Works
UV-Vis spectroscopy (or spectrophotometry) is a quantitative method used to measure the amount of light absorbed by a chemical. This is done by measuring the intensity of light transmitted through a sample (relative to the intensity of light transmitted through a blank sample). This method can be used for a variety of sample types, including liquids, solids, thin films, and glass.
Spectrophotometry is the quantitative measurement of the absorption/transmission or reflectance of a substance as a function of wavelength. Although it is called UV-Vis, the wavelength range typically used ranges from 190 nm to 1100 nm in the near infrared.
Using a spectrophotometer and making absorption/transmission measurements, we can determine the amount (or concentration) of a known chemical by studying the number of photons (light intensity) that reach the detector.
The greater the absorption of light by a substance at a specific wavelength, the greater the concentration of the known substance.
Applications of UV-Vis Spectrophotometer
UV/Vis spectroscopy is used in analytical chemistry for the quantitative determination of various analytes, including the transport of metal ions, organic compounds, and biological macromolecules.
